Proteome Analysis: 2-D Electrophoresis
GBS will perform two-dimensional electrophoresis (2DE) on sample material provided by the customer.* The first dimension is conducted with immobilized pH gradient (IPG) strips using a Amersham Biosciences Multiphor II system. The system used for the second dimension depends on which size gel format is required. For minigel format, a Biorad Protean III system is used; for standard format a Hoeffer 600 system is used, and for large format an Ettan Daltsix (Amerisham Biosciences) system is used. Customer may submit sample material ready for analysis or raw materials including tissues, tissue cultures, cell lines, bacteria, yeast and other microorganisms, and GBS will process the raw material for an additional fee. (Organisms requiring special procedures for cellular disruption have an additional fee).
If the customer wishes to conduct initial sample disruption in-house, GBS can mail its optimized denaturing buffer cocktail for final sample dilution for a minimal charge. If samples are provided in denaturing buffer cocktail (ours or the customers) the total protein value for each sample will need to be provided to ensure proper loading quantities.
If GBS does the sample disruption total protein will be calculated for an additional charge. For isoelectric focusing (first dimension) customers may choose from a variety of IPG strips of broad to very narrow pH ranges, and from four different strip lengths. For the second dimension, the customer may choose from several total acrylamide concentrations (range 8-18%) for gels with homogenous acrylamide percentages, or request custom gradient gels for an additional charge.
Additionally, customers may request 2DE under non-denaturing or non-reducing conditions in either dimension or both. Customer may choose from three different size formats, and several different total protein, and specialized protein staining methods. Results are returned as a JPEG image, with a description of the running conditions.
