
For customers choosing this service, the experts at GBS will apply
two-dimensional electrophoresis (2DE) to the protein material provided, whether processed or raw. Acceptable raw materials include tissues, tissue cultures, cell lines, bacteria, yeast and other microorganisms. When customer samples are provided in a denaturing buffer, the total protein value for each sample must be given to ensure proper loading quantities. If a customer conducts the initial sample disruption, GBS can mail an optimized denaturing buffer for final sample dilution at a minimal charge. Alternatively, GBS can perform specialized disruptions, general processing of raw material, and/or calculations of total protein for additional fees.
Note: If GBS does the sample disruption, total protein will be calculated, and there will be a charge for this computation.
For the 2DE service, the first dimension (isoelectric focusing or IEF) is conducted with immobilized pH gradient (IPG) strips using an
Amersham Biosciences Multiphor II system. Customers may choose from the following options:
The system used for the second dimension (SDS PAGE) depends on the gel size required. For the minigel format, a Biorad Protean III system is used; for standard format, a Hoeffer 600 system is used; and for large format, an Ettan Daltsix (Amersham Biosciences) system is used.
